Optimization of β-galactosidase production from lactic acid bacteria

چکیده
β-galactosidase, commonly known as lactase, represents commercially important enzyme that is prevalently used for lactose hydrolysis in milk and whey. To the date, it has been isolated from various sources. In this study different strains of lactic acid bacteria were assessed for their β-galactosidase productivity, and Lactobacillus acidophilus ATCC 4356 resulted with the highest production potential. Thereafter, optimal conditions for accomplishing high yields of β-galactosidase activity were determined. Maximal specific activity (1.01 IU mL) was accomplished after 2 days shake flask culture fermentation (150 rpm) at 37 °C, with modified Man Rogosa Sharpe culture broth using lactose (2.5%) as sole carbon source. Finally, in order to intensify release of intracellular β-galactosidase different mechanical and chemical methods were conducted. Nevertheless, vortexing with quartz sand (150 μm) as abrasive was proven to be the most efficient method of cell disruption. The optimum temperature of obtained β-galactosidase was 45 °C and the optimum range pH 6.5–7.5.

α-amylase and β-galactosidase production on Potato starch waste by Lactococcus lactis subsp lactis isolated from pickled yam
Potato starch waste, a chips industry effluent, was used for the production of industrially important enzymes by an amylolytic strain of lactic acid bacteria, isolated from pickled yam & identified as Lactococcus lactis. The strain was observed to co-produce α-amylase and β-galactosidase. Potato starch waste was efficiently utilized (91.6%) along with high co-production of α-amylase and β-galac...

متن کاملβ-galactosidase Production by Aspergillus niger ATCC 9142 Using Inexpensive Substrates in Solid-State Fermentation: Optimization by Orthogonal Arrays Design
Background: Enzymatic hydrolysis of lactose is one of the most important biotechnological processes in the food industry, which is accomplished by enzyme β-galactosidase (β-gal, β-D-galactoside galactohydrolase, EC 3.2.1.23), trivial called lactase. Orthogonal arrays design is an appropriate option for the optimization of biotechnological processes for the production of microbial...
متن کاملImpact of enzymatic hydrolyzed lactose on fermentation and growth of probiotic bacteria in whey
Whey is a green-yellowish fluid that derives from cheese-making and casein manufacture. The compositional quality and properties of whey can differ greatly, which depends on production technology and milk quality used. The main constituents of whey’s dry matter are lactose (46-52 g/L) and whey proteins (6-10 g/L) (Jelen, 2003; Adam et al., 2004; Jeličić et al., 2008). Whey fermentation is initi...
متن کاملA Study on the Antibacterial Activity of Lactic Acid Bacteria Isolated from Traditional Iranian Milk Samples
198 Lactic acid bacteria (LAB) were isolated from Iranian milk samples. Almost all isolatedLAB showed inhibitory activity towards E. coli and S. aureus as Gram negative and Grampositive indicators, respectively. No role was observed for hydrogen peroxide in the activity ofall isolated strains while organic acids proved a prominent role in all detected inhibitoryactivities. Within isolated LAB, ...
